The Triple Agonist: Retatrutide & the Next Frontier of Weight Loss

Retatrutide is Eli Lilly's experimental once-weekly injectable drug that targets three hormone receptors simultaneously — GLP-1, GIP, and glucagon — making it the first of its kind. In its first Phase 3 trial (TRIUMPH-4), patients on the highest dose lost an average of 28.7% of their body weight (roughly 71 pounds) over 68 weeks, while also seeing dramatic reductions in knee pain. These numbers surpass every weight-loss drug currently on the market, including tirzepatide (Zepbound) and semaglutide (Wegovy). Beyond weight loss, the drug improved blood pressure, cholesterol, and inflammation markers. A new side effect — abnormal skin sensation (dysesthesia) — emerged in Phase 3 and will be closely watched. Seven more Phase 3 trials are expected to report results throughout 2026, with FDA approval possible as early as 2027. Retatrutide is not yet available and remains investigational.

BPC-157: The Body's Own Healing Peptide — What the Science Actually Says

BPC-157 is a synthetic 15-amino-acid peptide derived from human gastric juice that has shown remarkable healing properties across 30 years of animal research — accelerating tendon repair, protecting the gut lining, reducing inflammation, and supporting neurological recovery. It works primarily by triggering new blood vessel growth, driving cell migration to injury sites, and modulating inflammatory cytokines without suppressing the healing process. The animal data is extensive and consistently positive. The human data, however, is nearly nonexistent — fewer than 30 people have ever been studied in published trials, a pivotal 2015 Phase 1 safety study was cancelled with no explanation, and no randomized controlled trials exist. The FDA classified it as a Category 2 substance in 2023, meaning it cannot be legally compounded. Despite this, it is widely sold online as a research chemical with zero quality control oversight. BPC-157 is one of the most scientifically interesting peptides in research today — and one of the least proven in humans.

Tesamorelin: The FDA-Approved Peptide Quietly Reshaping Metabolic Medicine

Tesamorelin is a synthetic 44-amino acid analogue of Growth Hormone-Releasing Hormone (GHRH), developed by Theratechnologies and FDA-approved since 2010 under the brand name EGRIFTA. It is the only peptide approved in the United States specifically to reduce excess visceral abdominal fat — the dangerous fat surrounding internal organs that drives cardiovascular disease, insulin resistance, and metabolic dysfunction. Unlike GLP-1 drugs that suppress appetite, tesamorelin works upstream at the brain-pituitary axis, restoring the body's own pulsatile growth hormone signaling rather than introducing exogenous hormones directly. Phase 3 trials in HIV lipodystrophy showed 15–19% visceral fat reduction at 26 weeks. A landmark Lancet HIV trial showed 37% relative liver fat reduction at 12 months, with fibrosis progression prevented. Emerging research points to applications in cognitive function, cardiovascular risk, and general metabolic disease. A simplified weekly-reconstitution formulation — EGRIFTA WR — was approved by the FDA in March 2025. In a space filled with unproven peptides, tesamorelin stands alone: it has the trials, the approval, and the documented safety data to back it up
